Promising New Drug Offers Hope for Pancreatic Cancer Patients

A new drug, daraxonrasib, is providing renewed hope for patients suffering from pancreatic cancer, a disease known for its grim prognosis. Clinical trials, including those at Penn Medicine’s Abramson Cancer Center, are showing promising results, particularly for patients like Irene Blair, who was diagnosed with stage-four pancreatic cancer in June 2023.

Blair, a 59-year-old grandmother from Newark, Delaware, was given a prognosis of only six to eight months to live. The advanced stage of her cancer, diagnosed less than a year earlier, left her with few options. However, participation in a clinical trial of daraxonrasib presented a potential lifeline.

Breakthrough Against a Deadly Cancer

Pancreatic cancer has one of the highest mortality rates among cancers, with only 13% of patients surviving five years post-diagnosis. The introduction of KRAS inhibitors, a class of drugs that block a protein responsible for cancer cell growth, has long been sought after. Daraxonrasib is a significant step forward, as it is one of the first KRAS inhibitors being tested for pancreatic cancer.

While daraxonrasib is not a cure, early clinical trial results are encouraging. In a phase 1 trial involving 38 patients, the drug appeared to double median survival time for half of the participants from approximately seven months to 15.6 months. Mark O’Hara, Blair’s oncologist and a leading researcher in KRAS inhibitors at Penn, emphasized the need for effective therapies beyond traditional chemotherapy.

Blair began her treatment in July 2023. Remarkably, within three weeks, her pain subsided, and her scans showed that her tumors had stabilized or decreased in size. By December 2023, follow-up imaging confirmed that her cancer had not progressed, marking a significant improvement from her prior chemotherapy regimen, which had left her weak and caused a weight loss of 35 pounds.

Impacts and Side Effects

While the drug is promising, it does come with side effects. The most common is a facial rash, which affected 91% of patients in the initial trial, with 8% experiencing severe cases. Other side effects can include nausea, diarrhea, and mouth sores, but O’Hara noted that these symptoms are generally manageable with medication. Importantly, many patients report a better quality of life on daraxonrasib compared to standard chemotherapy.

The federal government has expedited the review process for this innovative treatment, reflecting the urgency of finding effective options for pancreatic cancer. This is particularly relevant given the recent announcement by former Nebraska Senator Ben Sasse, who disclosed his own diagnosis of stage-four pancreatic cancer. His situation underscores the pressing need for advancements in treatment.

As Blair looks toward the future, she hopes to reclaim her life and enjoy time with family, including planned trips to California and Florida. “You just wonder, ‘Will I be here next year?’” she remarked, highlighting the emotional toll the disease takes on patients and their families.

Research into KRAS inhibitors has been ongoing since the protein’s discovery in 1982. With more than 90% of pancreatic cancer cases involving KRAS mutations, this class of drugs offers a targeted approach to treatment. The success of daraxonrasib may pave the way for further advancements in tackling this aggressive cancer.
