New Research Raises Concerns Over Gut Health Impact of Ozempic and Wegovy

Researchers at Adelaide University are examining the potential health implications of a lesser-known ingredient in the oral formulations of popular weight loss medications, Ozempic and Wegovy. The study, released on March 1, 2026, highlights the compound salcaprozate sodium (SNAC), which is critical for the absorption of semaglutide, the active ingredient in these medications. While the preliminary findings do not indicate harm to humans, they raise questions about the long-term effects of daily exposure to SNAC.

This research marks the first systematic investigation into how repeated exposure to SNAC may impact gut microbiota and related health markers. Conducted over a 21-day period using an animal model, the study observed significant alterations in gut bacteria and inflammation markers. Although it does not conclusively link SNAC to health risks in humans, the results suggest that its role may extend beyond merely facilitating drug absorption.

Understanding the Role of SNAC in Weight Loss Medications

Semaglutide, used in widely prescribed medications such as Ozempic and Wegovy, is designed to be injected for immediate absorption into the bloodstream. However, the introduction of a tablet version last year in the United States has increased interest in oral formulations. SNAC is essential in these tablets, providing a protective barrier against stomach enzymes that would otherwise degrade semaglutide. Without SNAC, the effectiveness of oral semaglutide would be compromised.

As the use of these medications rises globally, it is anticipated that daily exposure to SNAC will increase, leading to a need for further research into its long-term implications. Currently, around 890 million adults and 160 million children worldwide are affected by obesity, a condition linked to numerous health complications. The United States has the highest obesity rate among OECD countries, with 43% of individuals aged 15 and older classified as obese. Australia follows with 31%, which is above the OECD average of 25%.

Calls for Further Research on Long-term Effects

Lead author Amin Ariaee, a PhD candidate at Adelaide University, emphasized the importance of understanding SNAC’s complete impact as oral obesity treatments become more prevalent. “Obesity is a complex, chronic disease with serious health consequences,” Ariaee stated. “While these medicines are effective and assist many people, we need to comprehend what repeated, long-term exposure to all ingredients in the pill means for the body—not just the active drug.”

The study revealed that SNAC might be linked to shifts in potentially harmful gut bacteria and elevated inflammatory markers, as well as depletion of proteins related to cognitive function. These findings underscore the necessity for continued investigation into the implications of long-term use of weight loss medications that contain SNAC.

Despite the promising nature of these medications, Dr. Paul Joyce, a senior research fellow, cautioned against jumping to conclusions. He noted, “Importantly, our findings do not prove that SNAC causes harm in humans. However, they do show that the ingredient enabling these tablets to work may have adverse biological effects beyond drug absorption.”

As the popularity of these medications continues to grow, a comprehensive understanding of every component involved becomes crucial. The potential benefits of Ozempic and Wegovy must be weighed against the possible long-term health implications of their ingredients.
